The third string of a normal E9 tuning should be tuned open to G#, and then raised by the B pedal to A. Your title line says A#. Hopefully that's just a typo.
It don't mean a thing if it ain't got that swing.

If the pedal doesn't move the string at all, and you've tried adjusting the nylon nut, flip the guitar over. Move the pedal by hand and watch the linkage all the way to the changer. You'll probably spot the problem. It's possible the pull rod has disconnected from the bellcrank and needs to be reattached.
